DNA Oligonucleotides are short, single- and double-stranded synthetic DNA sequences that can be used in a variety of molecular biology applications. Using the Oligo Entry ordering tool, you can design your oligos to contain any DNA sequence you require and select from hundreds of modifications to suit your specific research needs.
Every DNA Oligo you receive will be deprotected and desalted to remove small molecule impurities. In addition, your oligos will be quality controlled via proprietary ESI-MS

An important measure of quality for any oligo manufacturer is coupling efficiency, because this directly affects the amount of full-length product you receive. Our manufacturing processes result in high coupling efficiencies (Figure 1) and are standardized across our global manufacturing sites (Figure 2) to provide you with most consistent quality in the industry. No matter where in the world you are conducting your research, an IDT oligo is an IDT oligo. Figure 1. IDT proprietary platforms have a better coupling efficiency than other suppliers, which provides more full-length oligonucleotides in your order. Small increases in coupling efficiency (≤1%) result in measurable increases in full-length product yield. The curves represent the theoretical yield for different lengths of oligos based on a coupling efficiency of 99.4% (IDT Oligos, n = 126) and 99.1% (other suppliers, n = 134 from three different suppliers) using the formula: percent full length product = (eff )(n-1)*100 where eff = coupling efficiency (for example, 99.4% = 0.994) and (n–1) is the number of coupling reactions needed to make an oligo of length n.

Many variables can lead to reduced oligo quality, including ambient temperature, humidity, and input reagents. Monitoring of coupling efficiency at IDT manufacturing sites ensures that we provide our customers with high-quality products. A quarterly average is shown (n = 16 per manufacturing site).

Frequently asked questions
What secondary structure considerations need to be included when designing primers for PCR?
When designing primers for PCR, two types of secondary structures should be analyzed: dimers and hairpins. For dimers, both self- and hetero-dimers should be reviewed. In general, the ΔG value for dimer analysis should be between 0 to −9 kcal/mol. Values more negative than this may adversely affect PCR reactions. For hairpins, the Tm of the hairpin should be lower than the annealing temperature for the reaction.
The Tm for the strongest hairpin should be at least 50°C below the annealing temperature. These secondary structures and their corresponding stabilities can be calculated online with the IDT SciTools™ OligoAnalyzer Tool.

How many reactions will I be able to do with 25 nmol PCR primers?
Most PCR reactions use 0.1−0.5 μM primer. Assuming a maximum concentration of 0.5 μM and a reaction volume of 20 μL, each reaction will require 10 pmol oligonucleotide primer.
For a typical 25mer oligonucleotide, 1 OD is equivalent to approximately 4 nmol, or 4000 pmol. Yield for a 20 base PCR primer on the 25 nmol scale is 12 nmol. Therefore, with even the minimum yield from a 25 nmol synthesis, you should be able to perform 1200 PCR reactions.
In summary: Minimum for a 20mer ≈ 12 nmol = 12,000 pmol = 1200 reactions.
